Population-specific variations of the genetic architecture of sex determination in wild European sea bass Dicentrarchus labrax L. ArchiMer
Faggion, Sara; Vandeputte, Marc; Chatain, Beatrice; Gagnaire, Pierre-alexandre; Allal, Francois.
Polygenic sex determination (PSD) may show variations in terms of genetic and environmental components between populations of fish species exposed/adapted to different environments. The European sea bass (Dicentrarchus labrax) is an interesting model, combining both a PSD system and a genetic subdivision into an Atlantic and a Mediterranean lineage, with genetic substructures within the Mediterranean Sea. Here, we produced experimental progeny crosses (N = 927) from broodstock sampled in four wild populations (North Atlantic, NAT; Western Mediterranean, WEM; North-Eastern Mediterranean, NEM; South-Eastern Mediterranean, SEM). We found less females than males in the progeny, both in the global dataset (32.5%) and within each paternal group (from 25.1% for...

Unfamiliar congener used as a visual attractor in wild caught and domesticated sea bass (Dicentrarchus labrax) placed in a T-maze ArchiMer
Benhaim, David; Begout, Marie-laure; Chatain, Beatrice.
The present work compared wild-caught and domesticated sea bass juveniles swimming activity, exploration and visual attraction induced by an unfamiliar congener located behind a transparent wall at the end of one arm of a T-maze. This cognitive challenge was based on the hypothesis that placed into a novel and therefore stressful environment; the fish would adopt a gregarious behaviour even though they were not familiar with the present congener. Twenty individuals of similar size from both origins were individually tested. After a 5min acclimatization period, the wall of the start-box was removed and the maze was filmed during 20 min. Different swimming variables including angular velocity (Vang), total distance travelled (Dtot), velocity mean (Vel), time...

Agonistic behaviour and feed efficiency in juvenile Nile tilapia Oreochromis niloticus ArchiMer
De Verdal, Hugues; O'Connell, Catherine Maeve; Mekkawy, Wagdy; Vandeputte, Marc; Chatain, Beatrice; Bégout, Marie-laure; Benzie, John A.h..
Given the strong effects of behavioural hierarchies on growth in many cultured species and the key role of feed efficiency in aquaculture economics, understanding the nature of the interaction of these variables is important for the sustainability of aquaculture. The relationship between agonistic behaviour, growth and feed efficiency in Nile tilapia, Oreochromis niloticus, was studied by rearing 120 fish in eight aquaria. Fish were video-recorded to estimate the occurrence of agonistic behaviour during a fasting and a refeeding period. Growth, feed intake and feed conversion efficiency (FCE - calculated as the inverse of feed conversion ratio (FCR)) were subsequently measured individually for each fish. Fish showed 58% less agonistic traits during the...

Impact of aquaculture on the genetic structure of Mediterranean populations of Dicentrarchus ArchiMer
Bahri Sfar, Lilia; Lemaire, Christophe; Chatain, Beatrice; Divanach, Pascal; Ben Hassine, Oum Kalthoum; Bonhomme, Francois.
A phylogeographic analysis based on 6 polymorphic microsatellites was performed on 15 sea bass samples. five from Western Mediterranean, seven from Eastern Mediterranean and three coming from French aquaculture stocks. Among the eastern samples, three did not cluster according to their geographic origin but rather with the occidental group. Furthermore. a somewhat lower allelic diversity was observed within these particular samples, indicating that they originated from a limited number of progenitors of foreign origin. Among the aquaculture stocks, only one revealed a significant reduction of genetic variability, indicating that these stocks are largely outbred and open to wild fishes. The fact that the use of occidental fingerlings to seed broodstocks in...

Response to domestication and selection for growth in the European sea bass (Dicentrarchus labrax) in separate and mixed tanks ArchiMer
Vandeputte, Marc; Dupont Nivet, M; Haffray, P; Chavanne, H; Cenadelli, S; Parati, K; Vidal, Marie-odile; Vergnet, Alain; Chatain, Beatrice.
Selective breeding of European sea bass (Dicentrarchus labrax) receives a growing interest, as the estimated heritability of growth is medium to high. In this study, we compared the offspring of four groups of sea bass sires, mated with the same wild dams: wild (W), first generation of domestication (D), first generation of mass selection for length (M), first generation of PROSPER-like selection for length (P). The comparison was done both in replicated tanks (separate rearing) and in mixed tanks (mixed rearing) where sire origins were recovered by genotyping of eight microsatellite markers. Weight, length and growth rate were measured from day 238 post-fertilization (69 g mean weight) to day 611 post-fertilization (390 g mean weight). Both in mixed and...

Relationship between individual and group learning in a marine teleost: A case study with sea bass under self-feeding conditions ArchiMer
Benhaim, David; Ferrari, Sebastien; Colchen, Tatiana; Chatain, Beatrice; Begout, Marie-laure.
Fish learning and cognition are usually approached by testing single individuals in various devices such as mazes that have serious drawbacks, especially in gregarious species, including the stress induced by the test procedure. This might impair the results and lead to misinterpretation about the learning abilities of the targeted species. In order to provide an alternative to the individual-based tests, we investigated for the first time the operant conditioning of four similar groups (50 individuals per tank) of sea bass. We used two computerized self-feeder devices per tank, each coupled with individual electronic identification and that were alternately activated during varying positive appetitive reinforcement period of time (7 to 1 day). Learning...

Can individual feed conversion ratio at commercial size be predicted from juvenile performance in individually reared Nile tilapia Oreochromis niloticus? ArchiMer
Rodde, Charles; Chatain, Beatrice; Vandeputte, Marc; Quoc Trinh, Trong; Benzie, John A.h.; De Verdal, Hugues.
Feed conversion ratio (FCR), the ratio between feed intake and body weight gain, is of major interest for improving aquaculture sustainability through reduced feed costs and environmental impacts. Demonstrating whether FCR measured in juvenile fish is an accurate predictor of their performance during the whole rearing period is critical to developing genetic improvement programs for this trait. This is especially true for estimates obtained in individually reared fish, for which this has high implications regarding the size of the necessary rearing structures. We obtained individual FCR from 30 male Nile tilapia Oreochromis niloticus from the GIFT strain individually reared in a recirculating system, from 36 to 260 g mean weight. They were fed twice a day...

Measuring individual feed efficiency and its correlations with performance traits in Nile tilapia, Oreochromis niloticus ArchiMer
De Verdal, Hugues; Mekkawy, Wagdy; Lind, Curtis E.; Vandeputte, Marc; Chatain, Beatrice; Benzie, John A. H..
Estimating individual feed intake of fish held in groups has long been a challenge precluding precise knowledge of the individual variation of feed efficiency (FE) in fish. In this study, counts of the number of feed pellets (1.63 mg on average) eaten by individual tilapia (Oreochromis niloticus) held in 8 mixed sex groups of 15 fish were measured from video recordings made over a period of 10 days where fish were fed twice daily to achieve compensatory growth after 10 days of fasting. The initial body weight of the fish was 9.77 ± 2.03 g. Accumulated measures of feed intake (FI) over 11 meals were found to achieve 95% repeatability and a high accuracy of estimation of FI. During the FI measurement period, the average fish growth was 12.0 ± 3.6 g, feed...

Genotype by diet interactions in European sea bass (Dicentrarchus labrax L.): Nutritional challenge with totally plant-based diets ArchiMer
Le Boucher, Richard; Vandeputte, Marc; Dupont-nivet, M.; Quillet, E.; Ruelle, Francois; Vergnet, Alain; Kaushik, Sadasivam; Allamellou, J. M.; Medale, F.; Chatain, Beatrice.
Aquaculture of carnivorous species has strongly relied on fish meal and fish oil for feed formulation; however, greater replacement by terrestrial plant-based products is occurring now. This rapid change in dietary environment has been a major revolution and has to be taken into consideration in breeding programs. The present study analyzes potential consequences of this nutritional tendency for selective breeding by estimating genetic parameters of BW and growth rates estimated by the thermal growth coefficient (TGC) over different periods with extremely different diets. European sea bass (Dicentrarchus labrax L.) from a factorial cross (1,526 fish) between 25 sires and 9 dams were used to estimate heritabilities and genotype by diet interaction. Starting...

Risk-taking behaviour variation over time in sea bass Dicentrarchus labrax: effects of day-night alternation, fish phenotypic characteristics and selection for growth ArchiMer
Millot, Sandie; Begout, Marie-laure; Chatain, Beatrice.
Differences in bold and shy personality on sea bass Dicentrarchus labrax were investigated between a population (wild) produced from wild-brood fish and a population (selected) produced from selected-brood fish. During the experiment (112 days), fish were reared under self-feeding condition to characterize the feeding behaviour of each individual fish. Three risk-taking tests (T1, T2 and T3 of 24 h with day-night alternation) were carried out at > 1 month intervals on 180 fish of each strain in order to monitor D. labrax behaviour over time and in relation to the light:dark period. A risk-taking score was evaluated via a preference choice between a safe zone (without food) and a risky zone (potentially with food) by recording the number and the duration...

A shortcut method for sexing juvenile European sea bass, Dicentrarchus labrax L. ArchiMer
Menu, Bruno; Peruzzi, Stefano; Vergnet, Alain; Vidal, Marie-odile; Chatain, Beatrice.
The objective of this study was to develop a rapid and effective method of sexing juvenile sea bass with minimum labour and material. To this end, the gonad squash mount technique was applied along with macroscopic techniques for sexing a large number of experimental fish at the age of 215-275 days post fertilization (p.f.). At this age, 90% of the 3894 fish could be unambiguously sexed by macroscopic examination of their gonads, whereas the remaining proportion was identified using squash mount preparations (10%). The accuracy of the observations was measured up to classical histology procedures. Undifferentiated fish accounted for 0.1% of the total population only. The application of the squash mount technique holds out the prospect of considerable...

Evaluation of behavioral changes induced by a first step of domestication or selection for growth in the European sea bass (Dicentrarchus labrax): A self-feeding approach under repeated acute stress ArchiMer
Millot, Sandie; Pean, Samuel; Leguay, Didier; Vergnet, Alain; Chatain, Beatrice; Begout, Marie-laure.
Among other strategies to improve fish welfare in rearing environment, domestication and/or selective breeding was proposed to minimize fish responsiveness to husbandry practices. To verify this hypothesis on a recently domesticated specie, the sea bass, Dicentrarchus labrax, L., an experiment was realized, using four populations differing according to their level of domestication or selection: one population produced from wild parents (Wild), one population produced from parents domesticated for one generation (Domesticated) and two produced from parents selected for growth for one generation (Selected A and Selected B). The experiment was carried out over 91 days with 600 fish (50 fish per tank, 150 fish per population). After a control period, the fish...

Estimation et amélioration des performances zootechniques de l'élevage larvaire de Dicentrarchus labrax et de Sparus auratus ArchiMer
Chatain, Beatrice.
By gathering specific measurement criteria and an objective analysis of field methods, the main purpose of this work is to help estimate larval Sea Bass (Dicentrarchus labrax) and Sea Bream {Sparus auratus) stock performance raised under intensive conditions. As such, it is a methodology guide zootechnicians can use, in their daily quantitative and qualitative evaluation stock issues. The first part of this work covers defining the precision of current counting methods and establishing their limitations. The quantification methods analysed, as well as the new proposed methods, relate to the major phases of larval farming, from laying to postlarval stage. Three field methods commonly used in aquaculture to quantify egg population are therefore...

Gene-centromere mapping in meiotic gynogenetic European seabass ArchiMer
Oral, Munevver; Colleter, Julie; Bekaert, Michhttps://w3.ifremer.fr/archimer-admin/author.jsp#ael; Taggart, John B.; Palaiokostas, Christos; Mcandrew, Brendan J.; Vandeputte, Marc; Chatain, Beatrice; Kuhl, Heiner; Reinhardt, Richard; Peruzzi, Stefano; Penman, David J..
Background Fully isogenic lines in fish can be developed using “mitotic” gynogenesis (suppression of first zygotic mitosis following inactivation of the sperm genome). However, genome-wide verification of the steps in this process has seldom been applied. We used ddRADseq to generate SNP markers in a meiotic gynogenetic family of European seabass (Dicentrarchus labrax): (i) to verify the lack of paternal contribution in a meiotic gynogenetic family; (ii) to generate a gene-centromere map from this family; (iii) to identify telomeric markers that could distinguish mitotic gynogenetics from meiotic gynogenetics, which sometimes arise spontaneously in mitotic gynogenetic families. Results From a single meiotic gynogenetic family consisting of 79 progeny, 42...

Heritability of Boldness and Hypoxia Avoidance in European Seabass, Dicentrarchus labrax ArchiMer
Ferrari, Sebastien; Horri, Khaled; Allal, Francois; Vergnet, Alain; Benhaim, David; Vandeputte, Marc; Chatain, Beatrice; Begout, Marie-laure.
To understand the genetic basis of coping style in European seabass, fish from a full factorial mating (10 females x 50 males) were reared in common garden and individually tagged. Individuals coping style was characterized through behavior tests at four different ages, categorizing fish into proactive or reactive: a hypoxia avoidance test (at 255 days post hatching, dph) and 3 risk-taking tests (at 276, 286 and 304 dph). We observed significant heritability of the coping style, higher for the average of risk-taking scores (h2 = 0.45 ± 0.14) than for the hypoxia avoidance test (h2 = 0.19 ± 0.10). The genetic correlations between the three risk-taking scores were very high (rA = 0.96–0.99) showing that although their repeatability was moderately high (rP =...

"Freezebass" Setting up of a sperm cryobank for sea bass. Q5CT-2002-71209. Final report (01 January 2003 – 30 June 2005) ArchiMer
Chatain, Beatrice; Poline, Olivier.
Objectives : Presently, most of marine fish farmers around the Mediterranean basin grow genetically wild fish strains, which have not been genetically improved for culture performance (growth, appearance, disease resistance etc.). It is inevitable that the present gap for genetically improved strains of marine fish, and their reliable supply, will be filled. It will ultimately dominate the competitive component of marine culture industries once they evolve into commodity-style agribusiness (with fish prices being set at the lowest possible cost of production and the best quality). Those countries, which can competitively develop and supply the improved strains, will enjoy the benefit (perhaps survival) for their local industries. They will avoid...

Negative genetic correlations between production traits and head or bony tissues in large all-female rainbow trout (Oncorhynchus mykiss) ArchiMer
Haffray, Pierrick; Bugeon, Jerome; Pincent, Cedric; Chapuis, Herve; Mazeiraud, Emmanuel; Rossignol, Marie-noelle; Chatain, Beatrice; Vandeputte, Marc; Dupont-nivet, Mathilde.
Genetic parameters of production traits (growth, carcass yield, fillet yield) and bony tissues (head and vertebral axis) were estimated for large all-female rainbow trout Oncorhynchus mykiss reared in freshwater. Genetic parameters were estimated using REML at 16 months of age (1636 g) on 1962 DNA-assigned progenies from a partial factorial mating design with 60 dams and 100 sex-reversed sires. Most traits presented medium to high heritability (0.37 to 0.54). A high genetic correlation (r(A)=0.97) was found between fillet yield and headless gutted carcass yield (or HGCarc%). Due to its higher heritability and high genetic correlation with fillet yield, selection on HGCarc% should result in a 50% increase in selection efficiency on fillet yield by reducing...
